Hybridization Chain Reaction Probe Generator
We are excited to venture into the realm of quick, easy, mutliplexable in situ hybridizations presented by the Hybridization Chain Reaction methodology (Choi et al. Development 2018). We wanted to make the technique more budget-friendly, have more control over probe design, and wanted to be able to report the exact sequences in publications for reproducibility purposes. As a result, we wrote this program that allows us far greater control and ease of ordering probes that mesh with the HCR system of reaction and amplification reagents.
The website tool is the easiest to use: HCR Probe Maker
You can also access the code on Github here.
Injection Mold 3D Printing File
We use a 3D-printed plastic mold for injecting Platynereis embryos. If you would like to get yours printed from Shapeways. (The listing belongs to the engineer Grant Harris we worked with.)
Here is also a handle for this mold. You will need to glue the two pieces together.
Here is a video explaining how to use this injection mold.
The images below show how the final mold should look like with the handle. And the pink arrow in the agarose image show the groove we use for injecting. All the other grooves were for testing purposes, and we typically do not use them.
Protocols
We do our best publishing extended protocols on GitHub along with each manuscript we submit. You can find them here.
